Ergothioneine: The 'Longevity Vitamin' Your Body Cannot Make on Its Own

L-Ergothioneine is a unique amino acid found in trace amounts in certain foods. New research suggests it plays a critical role in protecting cells from oxidative damage — and may be one of the most important dietary compounds for healthy aging.

What Is L-Ergothioneine?

L-Ergothioneine (ERGO) is a naturally occurring amino acid and thione antioxidant. It was first isolated from ergot fungus in 1909 and is found in small amounts in mushrooms, black beans, oat bran, and certain animal tissues. Crucially, humans cannot synthesise ergothioneine — we can only obtain it through diet or supplementation.

What makes ergothioneine scientifically remarkable is that the human body has evolved a dedicated transporter for it — the ergothioneine transporter (ETT), encoded by the gene SLC22A4. The existence of this specialised transporter, found in no other known dietary compound, strongly suggests that ergothioneine plays an important and specific biological role.

Targeted Cellular Protection

Unlike most antioxidants, which distribute broadly throughout the body, ergothioneine is actively concentrated in tissues with the highest metabolic activity and oxidative stress — including the liver, kidneys, bone marrow, eyes, and brain. Within cells, it accumulates specifically in mitochondria and nuclei: the two structures most vulnerable to oxidative damage.

This targeted distribution means ergothioneine provides antioxidant protection precisely where it is most needed — at the sites of energy production and genetic information storage.

The "Longevity Vitamin" Hypothesis

In 2018, Professor Bruce Ames — one of the most cited scientists in the world — proposed that ergothioneine should be classified as a "longevity vitamin." His hypothesis, published in PNAS, argued that ergothioneine deficiency accelerates the degenerative diseases of aging, and that adequate intake is essential for long-term health.

Supporting this hypothesis, epidemiological research has found that lower plasma ergothioneine levels are significantly associated with several health outcomes.

  • Cognitive decline and increased risk of mild cognitive impairment
  • Frailty and reduced physical function in older adults
  • Cardiovascular disease markers
  • Higher all-cause mortality in some population studies

Dietary Sources and the Supplementation Gap

The primary dietary source of ergothioneine is mushrooms — particularly shiitake, oyster, and king oyster varieties. However, modern diets — especially those low in mushrooms and organ meats — provide very little ergothioneine. Studies suggest that many adults in Western populations have ergothioneine levels well below what may be optimal for cellular protection.

DR. ERGO™ — the patented L-Ergothioneine in ErgoNMN™ — provides a standardised, bioavailable dose of ergothioneine in every capsule, bridging the gap between typical dietary intake and the levels associated with optimal cellular protection in research studies.
